=== Preventing and treating infection === To lower the bacterial count in wounds, therapists may use topical antibiotics, which kill bacteria and can also help by keeping the wound environment moist, which is important for speeding the healing of chronic wounds. Some researchers have experimented with the use of tea tree oil, an antibacterial agent which also has anti-inflammatory effects. Disinfectants are contraindicated because they damage tissues and delay wound contraction. Further, they are rendered ineffective by organic matter in wounds like blood and exudate and are thus not useful in open wounds. A greater amount of exudate and necrotic tissue in a wound increases likelihood of infection by serving as a medium for bacterial growth away from the host's defenses. Since bacteria thrive on dead tissue, wounds are often surgically debrided to remove the devitalized tissue. Debridement and drainage of wound fluid are an especially important part of the treatment for diabetic ulcers, which may create the need for amputation if infection gets out of control. Mechanical removal of bacteria and devitalized tissue is also the idea behind wound irrigation, which is accomplished using pulsed lavage. Removing necrotic or devitalized tissue is also the aim of maggot therapy, the intentional introduction by a health care practitioner of live, disinfected maggots into non-healing wounds. Maggots dissolve only necrotic, infected tissue; disinfect the wound by killing bacteria; and stimulate wound healing.

At some time after the Norman conquest of England in 1066 a motte-and-bailey was erected on the present site of Castle Park. Bristol was held by Geoffrey de Montbray, Bishop of Countances, one of the knights who accompanied William the Conqueror. William ordered stone castles to be built so it is likely that the first parts of Bristol Castle were built by Geoffrey in his reign. After the Conqueror's death (1087), Geoffrey joined the rebellion against William Rufus. Using Bristol as his headquarters, he burned Bath and ravaged Somerset before submitting to Rufus. He eventually returned to Normandy and died at Coutances in 1093. Rufus created the Honour of Gloucester, which included Bristol, from his mother Queen Matilda's estates and granted it to Robert Fitzhamon. Fitzhamon enlarged and strengthened Bristol castle and in the latter years of the 11th century conquered and subdued much of south and west Wales. His daughter Mabel was married in 1114 to Henry I's bastard son Robert of Caen. Her dowry was a large part of her father's Gloucestershire and Welsh estate and Robert of Caen became the first Earl of Gloucester, c. 1122. He is believed to have been responsible for completing Bristol castle. In 1135 Henry I died and the Earl of Gloucester rallied to the support of his sister Matilda against Stephen of Blois who had seized the throne on Henry's death. Stephen attempted to lay siege to Robert at Bristol in 1138 but gave up the attempt as the castle appeared impregnable.

== Function == YAP1 is a transcriptional co-activator and its proliferative and oncogenic activity is driven by its association with the TEAD family of transcription factors, which up-regulate genes that promote cell growth and inhibit apoptosis. Several other functional partners of YAP1 were identified, including RUNX, SMADs, p73, ErbB4, TP53BP2, LATS1/2, PTPN14, AMOTs, and ZO1/2. YAP1 and its close paralog, TAZ (WWTR1), are the main effectors of the Hippo tumor suppressor pathway. When the pathway is activated, YAP1 and TAZ are phosphorylated on a serine residue and sequestered in the cytoplasm by 14-3-3 proteins. When the Hippo pathway is not activated, YAP1/TAZ enter the nucleus and regulate gene expression. It is reported that several genes are regulated by YAP1, including Birc2, Birc5, connective tissue growth factor (CTGF), amphiregulin (AREG), Cyr61, Hoxa1 and Hoxc13. YAP/TAZ have also been shown to act as stiffness sensors, regulating mechanotransduction independently of the Hippo signalling cascade. As YAP and TAZ are transcriptional co-activators, they do not have DNA-binding domains. Instead, when inside the nucleus, they regulate gene expression through TEAD1-4 which are sequence-specific transcription factors that mediate the main transcriptional output of the Hippo pathway. The YAP/TAZ and TEAD interaction competitively inhibits and actively dissociates the TEAD/VGLL4 interaction which functions as a transcriptional repressor.
